New vid: Making and Using Agar

In this video I show how to make and use agar.  Thought it would be a good idea to put this vid out before publishing the Pan Cyan TEK vid which I've been working on for far too long (I know).  I want to give away lots of spore prints but only if people know what to do with a spore print ;) Hopefully this vid makes it clear.  This video is also on Rumble and on Odysee.  Note that making a spore syringe may be a better alternative for beginners (I created a video showing that as well)

Petri dishes

Inoculation loops

Xacto Knife  (for scraping spores and cutting agar)

Premixed MEA powder (not recommended but for the lazy)

To make your own mix: Light Malt Extract

Agar Agar Powder or this one

Lighters for flame sterilization

Good value blender or Blendtec

Large pressure cooker (great value and good reviews)  or smaller one

Kitchen Scale (great reviews, inexpensive)

Large plastic funnel with strainer insert

Optional parafilm but ziplock bags work fine.

Optional stuff only if making PDA or black agar: Honey and Activated Carbon

Don't forget to follow me on Patreon and request a free spore print if you haven't already received one and could use it.  My next video will NOT be on YouTube.

@gordotek I am brand new to mycology and I am finding your channel very interesting and helpful. I have couple questions. I recently purchased some spore siringes, my question is what is the different between spore siringes and culture siringes? If there is a difference what is the process to turn spores into culture? The second questions. by mistake I ordered a spore print instead of a spore siringe, I followed instructions on how to inoculate an agar petri dish with spores. I now have nice little round fluffy mycelium growths, but each has a greenish dot on the center. Is this normal or is any greenish stuff a bad thing?

Throwing my 2 cents of very little experience in trying to re-use unused agar. i've tried: 1. re-pressure-cook it 2. microwave it. Result = i could not get it to become somewhat liquid again. it remained gelatinous and therefore unusable. I'm remaking agar from scratch, just halving the ingredients.

@gordotek in your video you pour 2-3 dishes out of each pint jar. How long can those jars keep and still be viable, and what is your process for unused agar?

Dr. Anteater

@Timothy Kramer You can use an Instant Pot for both grain spawn and agar with no problem. For agar, transfer your media jar to your still air box or flow hood while still hot. If your sterile procedure is good, you'll have contamination free plates.

I have the products ordered and building a hood. Thanks for sending me a print and all your helpful instructions. I see you have a Kill-A-Watt connected to the heater. I use a Kill-A-Watt to testing how much juice a device is pulling, is that why you use it with the aquarium heater? Also I've used a Instant Pot to prep grain jars and have had success. It reaches a couple point short of 15PSI. Any thoughts on using a Instant Pot. I will do the agar in it and will let you know how it turns out. Should I Run blank plates to test? I have cube prints also I can run as a test before using your print.
