How about a mad scientist with a "chainsaw"? During the day of the 13th, a pumpkin spawns somewhere near the village, maybe near walls. Once you break that a message like "you can hear echoes of a chainsaw in the distance" appears, that would set the boss spawning during the night. At the night, the sky goes red and chainsaw sounds will appear at the same time as the boss. The boss could be one of the villagers with an open lab coat and nothing at the bottom. Maybe a half broken mask in wich you can see a big smile with pointy teeths and crazy eyes. The weapon sounds like a chainsaw but in reality is a two handed dragon dildo (maybe with 3 spikes that move up and down like a chainsaw). He then would poceed to attack the villagers by bonking them with the dildo, maybe give it a charge attack to close the distance between objectives. Also he may leave some candy mines on the floor from time to time. And throw explosives pumpkins!

Can it be a big momma dragon that turn your pups in to pumpkin so you have to use your walls to keep them safe. The minion are kobolds that lure pups or capture and offer them to the queen. Now the twist is that the dragon hate candy and you can put candy bowels for defense. Oh to summing her you eat candy and not share

You could make it on wave 666, but that might seem unplayable for some people or just switch the wave number to 666 temporarily when the boss appears. You could make it a sexy plague doctor (because that would be ironic and spooky at the same time). You wouldn't want to touch him/her given all the plague they have been around. A sexy ghost! Also ironic because he/she is untouchable. A sexy werewolf. This one is also ironic because he/she probably has flees. A sexy mummy! Ew, so old and gwoss. Who wraps themselves in that stuff anyways? You might also add some hidden npc's in the game that you can attract by building a totem or other item. I would love some ghosts or a dinosawr that says rawr hehe.
